More Info on Sharon D. Clarke's Doctor Who Character
Sharon D. Clarke appeared on BBC Radio 4's Loose Ends yesterday (relevant part at 6:34) and revealed that her character, Mary, on Doctor Who series 11 is a human and married to Bradley Walsh's character, Graham. This seems to rule out the Missy theory, barring some sort of disguise/dramatic reveal.
